The dynamic range of proteins in biological samples can reach twelve orders of magnitude in serum samples and more than 6 orders of magnitude in tissue fractions. Therefore it is necessary to pre-fractionate the samples before MS analysis. A successful pre-fractionation reduces the complexity of biological samples as well as ion suppression caused by highly-abundant proteins.

Fractionation can be performed on protein and peptide level where proteins are preferentially separated using gel-based techniques and peptides are separated gel-free using liquid chromatography. In proteomics approaches multidimensional pre-fractionation is used in different combinations like gel/LC, offgel electrophoresis/LC or LC/LC. For high throughput sample preparation we provide robotic assisted workflows.
