To immobilise molecules of interest onto a surface a coating is needed. For glass slides used as support in almost all microarray applications, this coating often consists of a simple silane bearing a reactive (often electrophilic) group for covalent binding DNA or proteins. For each application, the slide coating needs to be adapted with regard to binding chemistry and molecular properties of the probe to be immobilised.
A large number of coatings is available for many applications including the immobilisation of oligonucleotides, cDNA, RNA, proteins, antibodies, peptides, carbohydrates, and small molecules. Detailed information on surface chemistry is given in:
